There are three blood tests used in diagnosing diabetes. They may or may not be part of what you are calling a "regular blood test".
1. Casual plasma glucose > 200 mg/dl and additional symptoms of diabetes
2. Fasting plasma glucose (FPG) >126 mg/dl OR
3. Results of a 2-hour 75-g Oral Glucose Tolerance Test (OGTT) > 200 mg/dl
A Hemoglobin A1C test is used to monitor long term glucose levels and is used in some cases to indicate further testing is necessary but in and of itself it is NOT a basis for a diagnosis. An A1C reading > 6.1 is suggestive of diabetes but not enough evidence to diagnose such.

A fasting serum sugar will tell you what your blood sugar is. If you’ve been fasting more than 8 hours and it’s over 120, you may be diabetic. You can do this test yourself with a home testing kit, costs about $100.00. A less sure way is with the urine diagnostic stix you can pick up cheap at a drugstore, but I’ve never gotten reliable readings off of these. The sure way is to get a doctor to to the "a1c" test, again fasting. Anything under 7 is normal. Anything over 9 you are considered diabetic. In between, maybe…

A "fasting" blood test is usually a good indicator. That is when you go to the doctor if you haven’t eaten in the last 8-12 hours your sugar might be high.
Typically a doctor won’t rely on a single blood test. (You might have ignored the fasting requirement and had a cookie that morning.). He’ll suspect diabetes and suggest a second test to confirm results.
At the lab you would have to request a diabetes test. This is not a regular test. You can also have a random or fasting blood glucose test done at your pharmacy or clinic. This takes only seconds and can give you peace of mind while you wait on the lab result.
